Metformin: Restraining Nucleocytoplasmic Shuttling to Fight Cancer and Aging
In this issue of Cell, Wu et al. employed C. elegans and human cell experiments to identify a pathway through which metformin increases lifespan and inhibits growth. A key transcriptional target, ACAD10, is activated when metformin induces nuclear exclusion of the GTPase RagC, thereby inhibiting mTORC1 through an unexpected mechanism.
متن کاملNucleocytoplasmic Shuttling of Endocytic Proteins
Many cellular processes rely on the ordered assembly of macromolecular structures. Here, we uncover an unexpected link between two such processes, endocytosis and transcription. Many endocytic proteins, including eps15, epsin1, the clathrin assembly lymphoid myeloid leukemia (CALM), and alpha-adaptin, accumulate in the nucleus when nuclear export is inhibited. Endocytosis and nucleocytoplasmic ...
متن کاملMetformin, aging and cancer
cancers are associated with aging [1]. Metformin, a widely used antidiabetic drug, has been linked to a reduced cancer incidence in some retrospective, hypothesis-generating studies [2]. Since cancer and aging may share certain molecular processes, it is plausible that metformin may prevent cancer by acting on the aging process. Consistent with this idea, several studies report a life span exte...
